To express our target proteins, the phospholipase A2 (BBa_K4091024) and the gamma phospholipase inhibitor (BBa_K4091069, the part this page describes), we use the T7 promoter in our circuit to induceprotein expression by adding IPTG (Isopropyl β-d-1-thiogalactopyranoside) in the media. To confirm whether or not we could express our proteins the results were analyzed via SDS-PAGE. We did to the protein expression to perform the inhibitory assay and to quantify how the kinetics of this enzyme actualy works, however, we were unable to finalize our experiments due to the fact that our distribution kit did not arrive. We know that the inhibitory mechanism of this specific interaction follows competitive inhibitory kinetics, based on this, we perform some mathematical models to predict some outcomes, and if we had got our inhibitory assay data, we would have the equation that describes the behavior in which the substrate is consumed over time.
